Jimmy and Gretchen meet at the wedding of Vernon and Becca Barbara. Becca rejected Jimmy's marriage proposal two years earlier, and he's convinced she only invited him to throw her marriage to Vernon in his face, but he showed up anyway, determined to ruin her day as payback. Gretchen came as the guest of Becca's sister Lindsay, who is Gretchen's best friend and the enabler of her sex-drugs-and-rock'n'roll lifestyle. Gretchen is the publicist for a rap trio and lives in a chaotic apartment, while Jimmy, a writer, can support himself in upscale Silver Lake and drive a BMW despite having published only one novel, written during his post-breakup depression, that has sold little and been remaindered at the local bookstore. He supports himself by writing for glossy magazines, in addition to the tiny royalty checks that occasionally arrive in his mailbox.

The series was co-created by Louis C.K., Zach Galifianakis and Jonathan Krisel; Krisel is also the showrunner and director. Galifianakis stars in the dual lead role as Chip Baskets, a failed professional clown in Paris, who instead becomes a local rodeo clown in Bakersfield, California, and his twin brother, Dale Baskets. Galifianakis, C.K., M. Blair Breard, Dave Becky, Marc Gurvitz and Andrea Pett-Joseph serve as executive producers, with FX Productions as the production company.

Louie is loosely based on comedian Louis C.K.'s life, showing segments of him doing his stand-up routine onstage, and depicting his life offstage as a divorced father of two girls. Each episode features either two stories (which may or may not connect thematically) or a longer full-episode story (often consisting of numerous connected shorter pieces). The stories of all episodes revolve around Louie.

The stand-up in the show consists of original material recorded for the series and is usually shot from the stage rather than from the more traditional audience perspective. The pieces are interspersed with short clips of Louie's stand-up, usually performed in New York comedy clubs, mainly the Comedy Cellar and Carolines in Manhattan. Sometimes these comedy segments are integrated into the stories, whereas other times, they serve to book them with a loosely connected topic. In the first season, blunt, socially awkward conversations between Louie and his therapist are also shown occasionally.
